FORNISERT

(A Novel Ocular Insert for Controlled Delivery of Cyclosporine A)
FORNISERT is an innovative silicone-based ocular insert developed for the sustained and localized delivery of Cyclosporine A (CsA) directly to the subconjunctival fornix. This drug delivery system aims to overcome the limitations of conventional CsA administration by enhancing drug retention time, reducing dosing frequency, and ensuring more consistent therapeutic outcomes in ocular inflammatory and immune-mediated conditions.

Cumulative Release Profile

As illustrated in Figure 1, the release curve demonstrates the gradual, controlled release of CsA over time, maintaining concentrations within the therapeutic window and avoiding peaks that may cause toxicity or valleys that result in sub-therapeutic exposure.

Proof-of-Concept Clinical study

A pilot clinical trial demonstrated the effective placement and retention of FORNISERT in the conjunctival fornix. Key visual references include (Figure2):

  • White arrows: indicate the anatomical fornix of the patient.
  • Yellow arrows: show the physical placement of the FORNISERT insert.
  • Black arrows: identify the location of pterygium, one of the targeted pathological sites for CsA therapy.
